چکیده

The ionic basis of the resting potential and of the response to acetylcholine (ACh) has been investigated in the cell body membrane of the fast coxal depressor motor neurone in the metathoracic ganglion of the cockroach Periplaneta americana. By means of ion-sensitive microelectrodes, intracellular concentrations of three ion species were estimated (mmoll"): [K]j, 144±3; [Na]j, 9±1; [Cl~],, 7±1. The resting potential of continuously superfused cells was -75.6±1.9mV at 22C. A change in resting potential of 42.0±2.5mV accompanied a decade change in [K]o. Experiments with (10~ molP) ouabain, Na injection, low temperature (10°C) and non-superfused cells indicated the presence of an electrogenic sodium pump.

Voltage-independent Block of a Neuronal Nicottnic Acetylcholine Receptor by W-methyl Lycaconitine

TV-Methyl lycaconitine is the most effective low molecular weight antagonist reported for an insect neuronal nicotinic receptor. In the present study, the citrate salt of this neurotoxin from the plant Delphinium brownii was found to inhibit [H]-o^bungarotoxin binding to nerve cord extracts of the cockroach Periplaneta americana with a K, of l^x lO^ 'mol l" 1 . At a concentration of 1-Oxl0~moll...

SHORT COMMUNICATION A SULPHYDRYL REDUCING AGENT, DITfflOTHREITOL, MODIFIES AGONIST-NICOTINIC RECEPTOR INTERACTION IN AN IDENTIFIED INSECT NEURONE

Nicotinic acetycholine receptors are present at high density in the nervous system of insects (Sattelle, 1980). Studies on the structure of nicotinic acetylcholine receptors have shown a degree of homology between insect a-like subunits and a-subunits from other species (Bossy etal. 1988; Marshall etal. 1990; Sawruk etal. 1990a). The locust ar-subunit, when expressed in Xenopus oocytes, is able...

Actions of bicuculline on cell body and neuropilar membranes of identified insect neurones.

Bicuculline and its methochloride salt block inhibitory postsynaptic potentials recorded from giant interneurone 2 (GI 2) of the cockroach Periplaneta americana following stimulation of cercal nerve X, but fail to block the response to gamma-aminobutyric acid (GABA) when this neurotransmitter is ionophoresed or pressure-injected onto the fine branches of GI 2 within the neuropile of the termina...

L-glutamate Receptors on the Cell Body Membrane of an Identified Insect Motor Neurone

Current-clamp experiments on an identified neurone have demonstrated the presence of L-glutamate receptors in the insect central nervous system. The cell body of the fast coxal depressor motor neurone (Df) in the metathoracic ganglion of the cockroach Periplaneta americana exhibits a hyperpolarizing response to L-glutamate, accompanied by an increase in membrane conductance. The response is dep...

Actions of Cholinergic Pharmacological Agents on the Cell Body Membrane of the Fast Coxal Depressor Motoneurone of the Cockroach

The pharmacological properties of cholinergic receptors on the cell body membrane of the fast coxal depressor motoneurone (Df) of the cockroach (Periplaneta americana) have been investigated. Parallel dose-response curves were obtained for the depolarizing actions of four bath-applied agonists, with the following order of effectiveness: nicotine>acetylcholine (ACh), in the presence of 1-0X10~M ...
